3.8.2 Preparation of bacteria cell inoculum for immobilization
A potent cadmium resistant bacterium was inoculated in LB broth and incubated at 28oC with shaking for overnight. The bacterial culture was centrifuged at 6,000 rpm for 10 min, it was then washed twice with sterile 50 mM phosphate buffer (pH 7.0). The cell pellets were re-suspended in 50 mM phosphate buffer (pH 7.0) (Appendix B, No. 7). Cell density at OD600 was measured and adjusted to an OD600 of 2.0. Viable bacterial cells in suspension were enumerated by plate count technique.
