For studies that determined the fates of the organisms in
untreated apple juice, 0.1ml aliquots of acclimatized cell suspensions
were inoculated into 9.9ml juice after which 0.5ml of this suspension
were inoculated into 49.5 ml juice, introducing an initial population
of approximately 4e5 log CFU/ml. Apple juice suspensions were
incubated at 25 C, after which aliquots of 1 ml were obtained at
predetermined time intervals. The sampled juices were subjected to
serial 10-fold dilutions with PBS and surface plated on respective
growth media. Plated E. coli were incubated at 35 C for 24e48 h. All
yeast strains were surface plated onto PDA and incubated at 25 or
30 C for24e48 h prior to colony enumerations. Microbial analyses of
the suspending apple juice showed that background microflora was
not present; hence the PDA used to enumerate surviving yeast cells
was not supplemented with selective agents.
