can beusedasprecursorsfortheproductionofbiofuelsorbiochemicals.MicrobialbiosynthesisofFFAs
can beachievedbyintroducinganacyl–acyl carrierproteinthioesteraseinto Escherichiacoli. Inthis
study,theeffectofmetabolicmanipulationofFFAssynthesiscycle,hostgeneticbackgroundandcofactor
engineering onFFAsproductionusingglycerolasfeedstockswasinvestigated.Thehighestconcentra-
tion ofFFAsproducedbytheengineeredstainreached4.82g/Lwiththeyieldof29.55%(gFFAs/g
glycerol),about83%ofthemaximumtheoreticalpathwayvaluebythetypeIIfattyacidsynthesis
pathway.Inaddition,crudeglycerolfrombiodieselplantwasalsousedasfeedstockinthisstudy.The
FFAproductionwas3.53g/Lwithayieldof24.13%.Theyielddroppedslightlywhencrudeglycerolwas
used asacarbonsourceinsteadofpureglycerol,whileitstillcanreachabout68%ofthemaximum
theoretical pathwayyield.
