2.4. ESI-MS analysis of PSP toxins
The peaks expected as gonyautoxins (GTXs) and Nsulfocarbamoyl
toxins (C toxins) by HPLC-FLD werepartially purified for ESI-MS analysis from the extract of
the toxicity assay by successive treatment with activated
charcoal and a Bio-Gel P2 (Bio-Rad Laboratories) column
(Kotaki et al., 1981). ESI-MS was performed on a SSQ7000
mass spectrometer equipped with an atmospheric pressure
ion source, and an electrospray ionization (ESI) interface
(Finnigan MAT, CA, USA) was employed for detection.
The spray voltage was set at þ4.5 kV and the heated
capillary temperature was maintained at 250 8C. Full-scan
spectra were acquired in the positive ion mode over the
mass ranges of m/z 200–700 for GTXs, and in the negative
ion peak mode for C toxins over the same range. Sample
introduction was via a Rheodyne 7125 injector with a 25 ml
loop through a reversed phase column (Mitysil RR-18,
Kanto Chem.). The mobile phase consisted of distilled water
and acetonitrile (95:5, v/v) with 0.1% acetic acid.
