Prior to investigating the feasibility of SSF with pretreated Miscanthus, fermentation was conducted in synthetic medium at 42 C. Exponentially growing cells were harvested and transferred to 200 mL of the medium supplemented with 5% glucose. The initial cell density was adjusted to OD600 = 0.2 and the cells were cultured at 30 C and 42 C with shaking at 150 rpm. After samples were removed at 12 h intervals, cell growth and ethanol concentrations were determined by measuring the cell density at 600 nm, and by using gas chromatography equipped with an HP-INNOWaX 19091N-133 column.
