For the spread plate technique, agar
aseptically is poured into sterile Petrie
dishes and allowed to harden. A very
small amount of culture (0.1 – 0.2 ml) is
added to the surface of the hardened
agar. It is then spread across the agar
surface with a sterile glass rod. (We call
these rods, ‘hockey sticks’ in the US.)
These plates are also incubated upside
down.
