Enzymatic hydrolysisPretreated and untreated rice straws were subjected to enzy-matic hydrolysis. An amount of 3 g of substrate and 60 ml of 0.05 Msodium citrate buffer (pH 4.8) were mixed. Then 0.5 g/l sodiumazide (as an antibacterial agent) and the hydrolytic enzymes wereadded to the mixture. The enzyme loading was 30 FPU cellulaseand 60 IU -glucosidase per gram of dry substrate. After that, theflasks were closed with butyl rubber seals and aluminum caps andincubated for 72 h in a shaker incubator at 45◦C and 120 rpm. Liq-uid samples were periodically taken at various times during thehydrolysis, and the supernatant was separated from the residualsolid materials by centrifugation (3000 rpm for 15 min) and storedat −20◦C before sugar content analysis.
