Phosphatidyl serine (PS) translocation from the inner to outer
regions of plasma membrane is an indication of the onset of apoptosis.
Cell surface PS was detected by a PS binding protein annexin
V conjugated with Cy3 using the annexin V-Cy3 apoptosis detection
kit. The live untreated cells were stained with 6-CF and emitted
green fluorescence. MLL and cisplatin treated cells showed a
double staining with annexin V and 6-CF, emitting red and green
fluorescence, respectively. The percentage of annexin V stained
cells were calculated by counting the average number of cells
stained in different fields under microscope. Annexin V positive
cells were found to be 46% for MCF-7 and 15% for HCT-15 under
MLL treated condition and 41% and 35% for MCF-7 and HCT-15,
respectively under cisplatin treated condition
