supernatant containing 5 or 15 mg protein (corresponding to a cellobiase activity
of 0.28 or 0.84 CBU, respectively) was added, and the mixtures were incubated at
50C with shaking at 200 spm for 24 h. SSF was started by the addition of yeast
cell preparations containing equal amounts of S. cerevisiae INVSc1, either
expressing AaBGL1 or supplemented with Novozyme 188 (Sigma), a commercial
preparation of cellobiase from Aspergillus niger. Various amounts of yeast cells
representing approximately 2, 3 or 5 CBU of total cellobiase activity were tested.
The amounts of dry cell weight added to tubes containing converge-milled
Japanese cedar pretreated with 5 mg T. reesei protein were 10, 15 and 26 mg (26
OD600 units). For the tubes with 15 mg T reesei protein, 8, 15 and 29 mg (38 OD600
units) of dry cell weight were added. The tubes were sealed to create oxygenlimited
conditions and were incubated in a shaking incubator at 39C at 100 spm
for 12e72 h.
