free MS semi-solid medium for another 12 weeks for studying the residual effects of
PGRs. George et al. (2008) concluded that a successful PGRs treatment induced the
growth of several small shoots from each explant over 4-6 week periods after subculturing
in PGRs free media. Therefore, sub-culturing of explants onto PGRs-free media
may be required until the levels of cytokinin within the tissues are reduced. Number of
shoots per explant, number of leaves per shoot, stem height and root length were observed
and recorded before sub-culturing to the new medium.
The experiment was conducted as completely randomized design (CRD) with
5 replications per treatment and 3 shoots per replication. The experimental data were
analyzed using SPSS software (version 15.0). Duncan’s Multiple Range Test (DMRT)
was used to test differences among treatment means.
