Protocol for Transforming Soybean cotyledons using A. rhizogenes
(Subramanian et al., 2005)
The soybean seeds (variety Williams 82) were planted in an artificial soil mix (Superfine germinating mix, Fafard, New Jersey) and placed in a mist chamber. If a mist chamber is not available, the
seedlings need to be sprayed on a daily basis with a mist-sprayer. This helps dislodge the seed coat and results in high quality cotyledons. The cotyledons were ready to harvest 5 days after planting. Younger or older cotyledons may not yield good results.
