2.11. Substrate specificity
To determine the activity of the enzyme against different substrates,
four different polymers, namely carbohydrate glycogen,
a/b-cyclodextrin, pulullan and maltooligosaccharide (degree of
polymerisation, DP 4–8), were used. The above substrates were
prepared individually by dissolving 0.1% (w/w) in 100 mM sodium
acetate buffer (pH 5.0). Soluble starch (pre-gelatinised and acid
hydrolysed) 0.1% w/w) in 100 mM sodium acetate buffer at pH
5.0 was used as a reference substrate for enzyme activity
comparisons.
