For monographic double-fermentation, a mixture of 12 g of the seed powder, 0.24 g of Gluczyme AF6, 0.042 g of Cellulase A Amano 3, 0.042 mg of Hemicellulase Amano 90, 24 ml of distilled water was reacted for 24 h at 50°C, and the saccharified juice was obtained by centrifuging for 20 min at 6000 ×g. Aliquots of the juice were taken and measured for glucose and galactose contents by enzymatic methods (F-kit glucose, F-kit galactose, Roche Diagnostics K.K.).
