We have presented an analysis to measure the
Stokes polarization parameters in a fluorophotometer
and a spectrofluorophotometer. In order to measure
the Stokes parameters in a spectrofluorophotometer
the Q and b matrix elements of the diffraction
grating must be known. We have shown how these
matrix elements can be obtained using an unpolarized
light source. We have also presented a new
method to obtain the a and b matrix elements using
completely polarized light. This new method allows
their determination along with the G factor over the
entire useful spectral range of the instrument.
