2.3.5 Screening for useful metabolic abilities
2.3.5.1 Antibacterial testing
Antibacterial activity was assessed for the Original isolates only on 7H9, CZ and YEME media. Each isolate was stab-inoculated onto each of the three media using sterile toothpicks and incubated at 30°C for 14 days. The antibacterial activity was tested against a range of Gram-positive and Gram-negative bacteria: M. aurum A+, E. coli ATCC 25922, S. aureus ATCC 25923, and E. faecium (a Van A strain). Each test bacterium was grown in 5ml Luria-Bertani Broth (LB) (Sambrook et al., 1989) and incubated at 37°C with agitation for 24h (except for M. aurum, which required a longer incubation of 48h). Gram stains wereperformed on each culture before measuring the optical density (OD) at 600nm using a Beckman DU®530 Spectrophotometer.
