The specific activities of the cells were examined for the hydrolysis
of olive oil. E. coli (Calb/Tll) and E. coli (Calb-Tll) showed the
same specific activity: 0.36 U/mg cdw (cell dry weight), slightly
higher than the activity of E. coli (Tll) (0.35 U/mg cdw) and much
higher than that of E. coli (Calb) (0.03 U/mg cdw). The results indicate
that TLL plays a leading role for the hydrolysis of olive oil,
while CALB has difficulty to accept olive oil for the hydrolysis. Since
expressing of two enzymes in one or two plasmids gave no difference
in the whole cell activity, E. coli (Calb/Tll) with one plasmid
was chosen for further study.
