Since agarose is a gel-type and relatively hydrophilic matrix,
its interaction with solid substrates such as transparent polymers
or glass is weak. Hence, we spent some time on finding
an appropriate support for a stable coating of the agarose film.
The coating of agarose on such a substrate could improve, significantly,
its physical stability, and facilitate its handling and
storage. Microscope glass slides seemed to be an appropriate
substrate, but during the activation or dye coupling processes,
the agarose film was partially or completely detached from the
support. Finally, aminosilane-derived glass slides [12] were used
successfully. A thin film of agarose gel adhered to the substrate
and made a stable and transparent support (ACGS) which was
quite appropriate for the preparation of optical sensors.
