Detection of Coliphages
Coliphages were detected from concentrated water samples
using Escherichia coli K12 Hfr (ATCC) as the host bacterium
according to the double layer agar method. Briefly,
5 mL of Trypticase peptone semisolid agar (1%) containing
500 μL of K12 in exponential growth phase and 500
μL of concentrated water sample were poured onto Trypticase
peptone solid agar. Plates were incubated at 37°C
for 18 h and the coliphage plaques counted.
