2.6. Measurement of cytokine production
For the cytokine immunoassays, cells (1  106 cells/mL) were cultured in 6-well
plates, incubated overnight, and stimulated with LPS (1 lg/mL) for 24 h. Cell supernatants
were then collected, and the levels of TNF-a, IL-1b, and IL-6 were quantified
using ELISA kits (KOMA Bio-tech., Seoul, Korea) according to the manufacturer’s
instructions. In brief, 100 lL of biotinylated antibody reagent and the sample were
added to the 96-well plates, which were pre-coated with anti-mouse TNF-a, IL-1b,
or IL-6 antibody. The plates were covered and kept at room temperature for 2 h and
washed three times with a washing buffer. Then, 100 lL of tetramethylbenzidine
(TMB) substrate solution was added and developed at room temperature. The reaction
was quenched by adding 100 lL of TMB stop solution, and the absorbance of
the plates was read at 450 nm with a microplate reader (Molecular Devices, Sunnyvale,
CA, USA).
