hi to you both
this is a nice piece of work
but difficult if yuou have to say that the pathogen is less affected by NRAMP compared to Bth
is it worth trying to take out slide item 4 time course a bit longer behind 6hrs just in case it helps for Bps ? may not be technically feasible
also could try adding gamma to the experiment with Bps to see if it does anyhitng
re your question on slide item 3 i think the data is marginal about role of NO in the panel C for Bth CFU- it might be there but currently only one data point significant at 12 hours. i think you would need to devote a separate experiment to prove this point- repeating the experiment to see if reproducible. titrating in the AG to show it really does make a dose dependent effect on CFU- this would then convince the referee i think.
just to check in item 3 panel A is the y axis label really nitrile - should it be nitrite-? sorry if i am confused./wrong
i think it would be worth doing some cytokine studies here since you say NRAMP controls it and worth a try to see what happens
so worth a small manuscript i think after a bit more work but sorry i cant come up with any clearer/ more novel suggestions.
all the best
