Allele-specific primers corresponding to 12 kinds of SNP
in B. oleracea were designed according to different combinations
between mismatch base and mismatch site.
Optimization of melting temperature, primer length and
amplified products length were achieved using primer
program WebSNAPER (http://pga.mgh.harvard.edu/cgibin/
snap3/websnaper3.cgi) [23]. Primer sequences were
screened against B. oleracea genome repetitive sequences
to minimize mis-priming.
