Introduction
The EU Commission regulation No 2065/2001 of 22 October 2001 has laid down detailed rules for the application of Council Regulation (EC) No 104/2000 as regards informing consumers about fishery and aquaculture products. The information includes specification of the commercial designation and scientific name, method of production of a species (“caught” or “caught in freshwater” or “farmed” or “cultivated”) and the area in which it was caught. In the case of cultivated species, article 5 of the regulation indicates that a reference should be made to the country in which the product undergoes the final developmental stage.
Protection of high quality products and of their manufacture procedures is the intention behind the “Label of origin”. Ecolabelling is another type of label proposed for use on fishery products, but, unlike the compulsory labelling on information, this is a voluntary label introduced by a private company—Unilever—and by the World Wide Fund for Nature in 1996. According to the Organisation for Economic Co-operation and Development (OECD) “environmental labelling is a voluntary granting of labels by a private or public body in order to inform consumers and thereby promote consumer products which are determined to be environmentally more friendly than other functionally and competitively similar products” (FAO, 2000).
There is a clear trend in the international market towards labelling products with information about their composition and quality, which brings about the need to develop and standardize analytical methods to either confirm the information given by the label or to uncover fraud.
It must be noted that while there are several methods suitable for species identification in seafood, there is yet none accepted for the unequivocal determination of (i) the geographical origin, (ii) wild versus cultivated specimens, (iii) ecologically produced or (iv) the life-history of the product (the storage and processing conditions to which it was submitted). Yet, these are important data necessary to confirm the traceability documentation of the product and to detect deceptions.
Classical methods for species identification
Most foods are made up of a mixture of tissues and they usually contain all the components of cells in a more or less well preserved state, depending on the time elapsed after death and the processing to which the food product was submitted. This is not the case of purified components, such as oils, flour, essences as vanillin, sugar, etc.—that may be used in the food or as health-diet supplements.
Speciation analyses in foods that contain most cellular components, have been typically carried out by the analysis of the proteins or the DNA contained in the product (Sotelo, Piñeiro, Gallardo, & Pérez-Martı́n, 1993). Both types of techniques are simple and do not require lengthy or toxic protocols. They can be subdivided in two groups (Table 1): those that target many loci, and those that target one or a few loci.
