Observations The different Coloration of aerial and substratum mycelia and diffusible pigments at bottom of inoculated plates were observed (Galatenko et al, 1982). The seven isolates of actinomycetes were selected on the basis of mycelium colouration and diffusible pigments. The morphology of the filaments or mycelium was determined by light microscopy. The gram’s stain was used to determine positive and negative strain of actinomycetes with the help of Nikon photo micrographic unit at the magnification of 100 X.
upto 105 fold with serial dilution method. 1ml of the each dilutions (105 fold diluted sample) was spread on nutrient agar medium (NA, pH 7.2) and casein starch agar (CSA, pH 7.2) medium (Ellaiah et al, 1996) glycerol aspargine agar and yeast extract glycerol agar.
