The effect of addition of the commercial enzyme BGL (Novozyme-188) to Celluclast 1.5 L was also investigated.
A constant dose of BGL (10 U/g of substrate) was added to the reaction mixture to enhance the release of RS.
In all hydrolytic reactions involving BGL, ‘‘enzyme supplementing” refers to protein content per gram of the pretreated biomass.
For estimating the RS, 100-lL samples were taken from the reaction mixture at regular intervals and immediately heated to 90 C to denature the enzymes.
Samples were then cooled and centrifuged for 10 min at 8000 rpm.
The resulting supernatant was used for RS analysis, and the saccharification yield was calculated as described previously [17].
