Tobacco leaves were obtained from Cijambu village, district of Tanjungsari, Sumedang. Newly harvested tobacco leaves were sliced and air dried for one week and ground with a blender into powder. The powder was soaked with methanol (technical grade) for 72 hours in a glass jar. The solution was filtered with Whatman filter paper No. 41. Filtrates were evaporated using a rotary evaporator under reduced pressure to yield crude extracts. The crude extracts were kept under low temperature (-2oC) in a freezer until use.
