The extremely thermostable -amylas
The extremely thermostable -amylase, MJA1, was
cloned on the plasmid, pMW200 [30] and expressed in E.
coli. An isolated colony of transformed E. coli was cul-
tured in Luria–Bertani (LB) medium (tryptone, 10 g l
−1;
yeast extract, 5 g l
−1; NaCl, 5 g l
−1; pH 7.0) supplemented
with 100mg l
−1 ampicillin and transferred to 500ml of
Terr–Broth (tryptone, 12 g l
−1; yeast extract, 24 g l
−1; glyc-
erol, 4 g l
−1;KH2PO4, 2.31 g l
−1;K2HPO4, 12.54 g l
−1;pH
7.4) supplemented with 100mg l
−1 ampicillin. The culture
was grown at 37 ◦C in a rotary shaker (200 rpm) for 15 h.
The resulting biomass was harvested by centrifugation at
6000 × g (20 ◦C) for 30min. The pellet obtained from
centrifugation was suspended in sodium phosphate buffer
(0.02M; pH 7) and disintegrated using a 550 Sonic Dis-
membrator (Fisher Scientific). Disruption was performed
at 0 ◦C for 5min with 1min intervals after each 1min ex-
posure. After removal of cell debris by centrifugation at
15 000 × g for 30min, the protein extract was heated at
100 ◦C for 15min, centrifuged at 15 000 × g for 10min,
and filtered through a 0.2m pore-size filter (Millipore,
Bedford, MA, USA). The cell-free extract was stored at
−20 ◦C until further use.
