2.5.2. Assessment of morphological alterations by scanning electron microscopy (SEM)
An overnight culture of S. aureus was centrifuged (4000 rpm, 10 min) and washed two times in PBS buffer. One percent (v/v) of culture inoculum (1 × 108 CFU/mL) was treated with 10-fold diluted cinnamon leaf oil based microemulsion CMF4 for 60 min. One drop of interacted and non-interacted (control) sample were coated onto glass slides and allowed to get dried. Specimens were then coated with gold by using a sputter coater. Processed samples were observed under a high resolution scanning electron microscope (FEI Quanta FEG 200) with a resolution of 1.2 nm gold particle separations on a carbon substrate.
