For pour plates, culture broth is aseptically
added to a sterile container called a Petrie
dish (Appendix 10). These may be made
of either glass or plastic. Then a small
amount (1 ml) of agar media at about
45 oC is added to the Petrie dish and the
dish gently agitated to mix media and
culture. Allow the Agar to harden.
Incubate the plates upside down at the
desired temperature for the time required.
