In vitro test for eicosanoid synthesis inhibition
Several methods of measuring levels of these compounds are
used but the use of a radioimmunoassay has been accepted by
many workers and the method used was identical to that used
for assessing the activity of the crude extracts of the plant [1],
[8]. Test extracts and compounds were used to give final concen-
trations in the range 10 ± 500 mg/mL for the extract and fractions
and 31.5 to 250 mg/mL for 1 and 2 and 1 to 50 mg/mL for 3 and 4.
All determinations were carried out in triplicate and indometha-
cin 5 mM was used as positive control for COX inhibition and the
methoxyalkylthiazole ZM-211965 5 mM for 5-lipoxygenase inhi-
bition.
