Trials were conducted to assess the effectiveness and sensitivity
of the technique. The immunogen used to generate the
monoclonal antibody was UV irradiated salmon sperm DNA. To
create a positive control, purified salmon sperm DNA was irradiated
for 15 min at 10 cm distance from a transilluminator (Model
UV-21, mid (312 nm) UV bulb, Fotodyne Inc, Hartland WI). Similarly,
purified DNA from F. candida was irradiated and both samples
examined for the presence of thymine dimers with non-irradiated
DNA present for negative controls.
