Pour plate method was used to determine the total yeast and
mold counts. Known amount (39 g) of potato dextrose agar (PDA)
powder was dissolved in 1000 mL of distilled water to prepare
media. To avoid the cross contamination, tartaric acid solution
(10:90, w/v) was added in the PDA. All the petri dishes containing
PDA were placed in an incubator at 321
C. Then after two days the yeast and mold were counted in each petri dish and the results
were shown as CFU/mL of juice. All the determinations were carried
out in triplicate.
