2.6. Culture of S. enteritidis
S. enteritidis was cultured overnight in a medium containing
1.0% polypeptone, 0.5% yeast extract, 0.3% glucose, 1.0% NaCl, and
0.1% MgSO4 at pH 7.0 and 37 C. Bacteria cells were harvested by
centrifugation at 5000g, at 4C for 10 min, washed with sterile
10 mM PB (pH 7.2), containing 0.15 M NaCl, and then suspended
in the same buffer solution. The bacterial density, expressed as
cfu/ml, was estimated using a standard curve of S. enteritidis cfu/
ml and absorbance at 600 nm.
