In the plants of the genus Fragaria, few chromosome images
had been obtained after staining with DAPI and lacto-propionic
orcein at pro-metaphase. Only Lim (2000) reported staining
the chromosomes of F. vesca with DAPI, but at the metaphase
stage. In addition, the pro-metaphase chromosomes of strawberry
plants had readily distinguishable shapes; counting
chromosomes after DAPI staining was feasible because the
chromosomes stained using the DAPI solution gave a stronger
reflection and brighter signals (Lim et al., 1998). The DAPI
staining methods described herein are inferred to be effective
for chromosome observation in strawberry plants.
