Isolation and Culture
Branchiomyces is not a fastidious organism. It
has been cultured on a variety of routine agar
media including Saubouraud's dextrose agar,
peptone-dextrose agar, and corn meal agar, as
well as a media consisting of 10% duck excrement
decoction, 10% gelatin, and 0.1% citric
acid that has been adjusted to a pH of 5.8. 3,5,9,69
The excised gill tissue (which may be rinsed in
an antibiotic solution to reduce contaminating
bacteria) is placed on the agar media and incubated
at 25~ to 30~ 9'69 The fungus appears to
grow better at warmer temperatures, and it is
sometimes difficult to induce reproductive structures
in culture.
