3.2. Effect of cytokinin type and concentration on shoot proliferation
Stem nodal explants were selected because of their high potential for inducing shoots as observed in the previous experiment. Three cytokinins (i.e. kinetin, BA and TDZ) at two different concentrations (i.e. 0.5 and 1 mg l−1) were tested for their influence on growth and shoot formation from stem nodal explants. Explant growth was observed in all cytokinin treatments for all three cultivars ( Table 2). No significant difference in growth between the different cytokinin treatments was observed for ‘Imperial Red’ and ‘Imperial Rainbow’, but Kn and BA treatments gave significantly higher growth response than the control treatment in the case of ‘Imperial Green’.
