Determined the Michaelis–Menten constant (KM) and the maximal reaction velocities (Vmax) of pectinase.
Incubated the homogenate in an appropriate buffer (pH 6) at 45 °C with the substrate pectin at concentrations ranging from 1.32 to 21.12 mg•mL−1.
Performed the experiments in triplicate.
Evaluated the KM and Vmax by nonlinear regression analysis using Sigmaplot 10.0 software.
