reduced (data not shown). In the subsequent fermentation, the pHlevel set to 7 was held with base addition, and growth was sustaineduntil beginning of stationary phase. Next, batch with defined mediawas performed for determination of heterologous AMP productionwith pH set to 7 and without dO2 level control. During exponentialphase in this condition, a drop in oxygen levels was accompaniedby an increase in acetate production (Fig. 4A). In order to lower theacetate level, dO2 was set to 20% and acetate production was minimized. Moreover, a 10% increase in total biomass was observed(Fig. 4B). To further increase the biomass production, in order toobtain a higher protein amount, a fed batch was done, with fed inpulses. In this condition, final biomass production was increased by18% (Fig. 4C). Analysis of protein yield (Table 1) indicates a higherproduction in TB media, of 11 mg of Pa-MAP 2 per gram of biomass(YP/X). In defined media, higher protein production was observedboth in batch and fed-batch strategy with oxygen control, yielding3.7 mg of Pa-MAP 2 per gram of glucose. Nevertheless, total Pa-MAP 2 production was higher in fed-batch strategy, with 96 mg L−1against 78 mg L−1in batch strategy.
