2.1.
Isolation
and
purification
of
polysaccharides
The
dried
fruiting
bodies
of
Jinqian
mushroom
were
pulver-
ized
by a miller.
The
power
(100
g)
went
through
a 60-mesh
screen
and
defatted
with
ethanol.
The
residues
were
dried
at 45◦C
and
extracted
with
hot
distilled
water
for
3 h at
90◦C
to obtain
water-soluble
polysaccharide
in yield
of
11.34%.
Subsequently,
the
resulting
materials
were
extracted
with
5 L of
1.25
M
NaOH/0.05%
NaBH4solution
twice
at
25◦C for
2 h.
The
combined
supernatants
were
neutralized
by
acetic
acid,
centrifuged
to remove
precipita-
tion,
concentrated
and
removed
proteins
by
Sevag
methods.
Then,
the
acetone
was
added
slowly
to the
supernatants
until
the
acetone
concentration
(v/v)
reached
50%
volume.
The
precipitations
were
obtained by centrifugation
(8000
rpm/min
at 50◦C
for
10 min),
thoroughly
dialyzed
(MWcutoff
3500)
and
lyophilized
to give
the
polysaccharide
fraction
JQPs
(Fig.
1b,
yield
of
9.78%).
