study, Enterobacter cloacae strain SDMwassystematicallyandmetabolicallyengineeredtoconstructan
efficient biocatalystforproductionofthefuelandenantiopurebio-chemical—(2R,3R)-2,3-BD. First,the
various(2R,3R)-2,3-BD dehydrogenaseencodinggeneswereexpressedina meso-2,3-BD dehydrogenase
encoding genedisrupted E. cloacae strain undernativepromoter Pb of the2,3-BDbiosyntheticgene
cluster of E. cloacae. Then,carboncataboliterepressionwaseliminatedviainactivationoftheglucose
transporterencodinggene ptsG and overexpressionofagalactosepermeaseencodinggene galP. The
resultant straincouldutilizeglucoseandxylosesimultaneously.Toimprovetheefficiency of(2R,3R)-2,3-
BD production,thebyproduct-producinggenes(ldh and frdA) wereknockedout,therebyenhancingthe
yield of(2R,3R)-2,3-BD by16.5%in500-mLErlenmeyer flasks.Byusingfed-batchfermentationina5-L
bioreactor,152.0g/L(2R,3R)-2,3-BD (purity497.5%)wasproducedwithin44hwithaspecific produc-
tivity of3.5g/[Lh]andayieldof97.7%fromamixtureofglucoseandxylose,twomajorcarbohydrate
components inlignocellulosichydrolysates.Inaddition,whenalignocellulosichydrolysatewasusedas
the substrate,119.4g/L(2R,3R)-2,3-BD (purity496.0%) wasproducedwithin51hwithaproductivityof
