Cell Viability Assay (MTT Assay). Cytotoxicity studies
can be performed in 96-well plates. The mechanically
scraped THP-1 cells will be plated at 1 × 105/well in 96-
well plates containing 100 μL of DMEM with 10% heatinactivated
FBS and incubated for overnight. Test samples
will be dissolved in DMSO (at 0.1% concentrations). After
overnight incubation, the test material will be added, and the
plates will be incubated for 24 h. Cells will be washed onceand added with 50 μL of FBS-free medium containing MTT
(5 mg/mL). After 4 h of incubation at 37◦C, the medium will
be discarded and the formazan blue that formed in the cells
will be dissolved in DMSO (100 μL), and the optical density
will be measured at 540 nm.
