Primary explants 60 to 90 d after induction became blackishbrown
in color. However, these primary explants could be used
for second and third rounds of somatic embryo formation in
fresh medium after removal of the embryos. This demonstrated
that blackish-brown colored explants are living and viable, and
harbor sufficient embryogenic potential. However, the number
of embryos formed was reduced from about 347 embryos per
explant in the primary harvest to 50 embryos per explant in the
subsequent harvest. To the best of our knowledge, the repeated
use of the primary explants for embryo formation has not been
reported in any other plant species.
