Ten grams of minced tuna meat were taken from each sterile
polyethylene bag and homogenized at high-speed for 2 min in
a sterile blender (Osterizer) with 90 ml of 0.1% peptone water. The
homogenatewas serially diluted with 0.1% peptonewater and 0.1ml
aliquots of the diluted sample were plated on aerobic plate count
(APC) agar (Difco) containing 0.5% NaCl in duplicate. Bacterial colonies
were counted after the plates were incubated at 35 C for 2 d.
