The effects of amino acids and fed-batch on glutathione (GSH)
production by Saccharomyces cerevisiae ATCC 7754 were investigated.
The fermentations were carried out in a 5 L fermentor in
order to obtain the best conditions for feeding the carbon sources
(sugar cane molasses, glycerol or glucose) and amino acids addition.
Among the trials, the highest GSH concentration was obtained
without feed and amino acids addition in 96 h (241.3 mg/L). In trials
with carbon sources feeding and amino acids addition, were
obtained lower results than the trials without feeding, which was
not expected. In studies with fed-batch, the highest GSH concentration
was 166.92 mg/L at 84 h. To improve the production of GSH in
fed-batch processes is suggested to keep the specific growth rate at
low values, the oxygen dissolved at high rates and monitoring the
ethanol concentration in the broth. In trials with amino acids addition,
these were added in different concentrations and times. The
highest GSH production was obtained at 84 h (236.02 mg/L) with
the addition of three amino acids in 32 h, whereas in the trial which
amino acids were added in 6, 24, 48 and 60 h, the maximum GSH
obtained was 179.45 mg/L. These results indicates that the addition
time and concentration of each of the three amino acids are
key aspects in the increase of GSH and should be better studied to
obtain better results.
