The RKN populations of M. incognita (isolated from greenhouses
in Shouguang, Shandong province, China) were maintained on
susceptible tomato plants, Lycopersicon esculentum cv. Jiafen-19.
These plants were grown in plastic pots with heat sterilized soil
in a greenhouse. The tomato roots were washed to remove the soil
and egg masses were separated from infected root parts after
infecting with nematodes for six weeks. Removed egg masses were
placed on a sieve and the J2 nematodes that passed the sieve were
cultured for 72 h at room temperature and collected as stock
cultures.
