2.7. Survival of E. coli O157 EDL-932 and L. monocytogenes Scott A in table
olives fermented by oleuropeinolytic starter culture (OSC) in reduced salt
brines and industrial elaboration
2.7.1. In olive homogenate
Fromeach fermentation vessel amounts of 50 g aseptically pitted ol-
ives were transferred under sterile conditions in stomacher filter bags
and after addition of 250 ml sterile distilled water, homogenized for
60 s in stomacher and transferred to sterile flasks. Each flask was inoculated
with E. coli O157 EDL-932 or L. monocytogenes Scott A at a concentration
of 1 × 10
7
CFU/ml or 1 × 10
6
CFU/ml, respectively.
After inoculation and after 30 min and 2, 4, 6, 8, 10 and 24 h of incu-
bation at 30 °C with shaking (100 rpm) the counts of E. coli O157 EDL932
or L. monocytogenes Scott A were enumerated. LAB counts were
enumerated before inoculation with foodborne pathogens and 24 h
after incubation at 30 °C.
Olive homogenates were serially diluted in peptone/saline diluent
and surface plated on MRS agar for enumeration of LAB incubated anaerobically
(GasPak, BBL, Cockeysville, Maryland, USA) at 30 °C for
2–3 days and on Sorbitol MacConkey agar for enumeration of E. coli incubated
at 37 °C for 24 h or on PALCAM agar for enumeration of
L. monocytogenes incubated at 37 °C for 24–48 h. When less than
100 CFU/ml of L. monocytogenes or E. coli were expected the most probable
number (MPN) technique (3 × 3, i.e. 3 × 10 ml, 3 × 1 ml, 3 × 0.1 ml)
was applied as reported below. In the case of E. coli, mediumE. coli with
novobiocin (mEC) broth tubes were incubated at 37 °C for 18–24 h and
turbid tubes were confirmed by plating on Sorbitol MacConkey agar
plus MUG (Sharlau). In the case of L. monocytogenes, Listeria enrichment
broth (Scharlau) tubes were incubated at 37 °C for 24 and 48 h. Positive
result (turbid tubes) was confirmed by streaking on PALCAM agar
(Scharlau).
