2.3.8. Tocopherol
The tocopherol content was determined according to the AOCS official method (1992), using an Agilent 1200 series HPLC equipped with a diode array detector. Two grams of B&D extract were evaporated under nitrogen and redissolved in 2 mL of hexane with 20 mL of tocopherol acetate as internal standard. An aliquot (10 mL) was injected onto a normal phase column (250 mm 4.6 mm 5 mm) (Teknokroma Analítica S.A, Barcelona, Spain). Elution was performed with an isocratic mixture of hexane:2-propanol (99.6:0.4; v:v) at a flow 1.3 mL/min. Detection was set at 295 nm and 284 nm for tocopherol acetate. Results were expressed in mg tocopherol/g muscle.
