Total starch was determined by the method of Al-Rabadi,
Gilbert, and Gidley (2009). Sample (50 mg) was initially taken
and starch was first broken into dextrins and maltose using thermostable a-amylase & further amyloglucosidase (Sigma) was
added to release glucose. Finally, the released glucose was estimated by GOD-POD reagent (Erba Mannheim, Transasia Biomedical Ltd., India) and total starch was expressed in g/100 g using
the formula:

where, DA is the absorbance read against the reagent blank; F is the
factor for conversion from absorbance to lg; W is the weight of
sample in mg; FV is the final volume.
The analysis was performed in triplicates.
