Marker assisted selection (MAS) was conducted to identify
individual plants carrying the donor alleles in the four backcross
generations and the BC4F2 population. Two markers were found to
be highly polymorphic between the parents. Functional Aromaker
distinguished a specific allele between Manawthukha (400 bp) and
Basmati (392 bp). For the microsatellite markers RM00 and
RM204, polymorphisms were identified between alleles based
on the number of simple sequence repeats, two kinds of DNA
profiles ranging from 310 bp and 325 bps and 105 bp and 124 bp
between Manawthukha and Basmati in RM00 and RM204,
respectively. These markers are co-dominant markers that can
identify the presence of heterozygous alleles derived from the
donor in all BCnF1 generations. Twelve plants possessing homozygous
alleles of the three markers were selected from BC4F2
generation These BC4F2 lines were self-pollinated to generate
BC4F3 and then used as introgression lines (IL) for the evaluation of
cooking quality and yield performance under target environments.
