The binding pose of compounds 1 and 6 are the closest ones, presenting the carbonyl group from the coumarin ring pointing towards the Asn297 (2.83 Å and 3.26Å, respectively). However, these compounds interact with the ferryl heme of the CYP2A6 in different ways: compound 6 uses the position 10 (2.98 Å), while the compound 1 interacts directly through the oxygen from the furan ring, similarly to what was observed for the potent xanthotoxin inhibitor (illustrated on the right side of Fig. 1).
