Effect of carbon source on cell density Glycerol was a superior carbon source to glucose (Fig. 1). High cell densities can be achieved relatively easily
using glycerol, while high concentrations of glucose and high growth rates of cells can result in unbalance between glycolysis and oxidation of metabolic
by-products. This leads to accumulation of by-products such as acetate in large quantities and this has been reported to have detrimental effects on recombinant cells [3]. On the other hand, the lower rate of transport
of glycerol into the cell, compared to that of glucose, appears to lead to a reduction in the flux of carbon through glycolysis. This greatly reduces the formation of growth-inhibitory by-products so that the rapid growth of recombinant Escherichia coli is inhibited [1]. The following equations can be employed to describe the relationship between cell density (X,g/l) and fermentation time (t,h) for the two carbon sources:
for glucose X = - 0.1371 t 2 + 9.0640t -- 15.9089,
r = 0.9899, (1)
for glycerol X = - 0.4929t 2 + 19.8843t - 29.36,
r=0.9934. (2)
The fitting results are satisfactory (Fig. 1). According to eqn (1) and (2), the cell density can bc expected to achieve the maximum value Xm = 133.9016 and 171.1804 g/l at the time to, = 33.0562 and 20.177 h for glucose and glycerol as carbon sources, respectively.
